EZ DNA Methylation-Gold

EZ DNA Methylation-Gold Kit는 기존 EZ DNA Methylation Kit를 개선한 제품으로, DNA Denaturation과 Bisulfite Conversion 과정을 하나의 단계로 통합하여 보다 빠르고 효율적인 Bisulfite 처리 Workflow를 제공합니다. 본 제품은 기존 EZ DNA Methylation Kit에서 사용되던 Chemical Denaturation 방식 대신 Heat Denaturation 방식을 적용하여, DNA 변성과 Bisulfite Conversion 단계를 동시에 수행할 수 있도록 설계되었습니다. 이를 통해 전체 Incubation Time을 크게 단축하면서도 간편한 실험 과정을 제공합니다. 또한 EZ DNA Methylation-Gold Kit는 Bisulfite 처리 이후 높은 수율의 DNA Recovery가 가능하도록 최적화되어 있으며, 처리 과정 중 DNA 손실을 최소화할 수 있습니다. 회수된 Bisulfite-Converted DNA는 높은 품질을 유지하며, PCR Amplification, Endonuclease Digestion, Sequencing, Microarray 분석 등을 비롯한 다양한 Downstream Analysis에 적합합니다.

Streamlined Workflow

정제된 DNA는 CT-Conversion Reagent에 직접 첨가하여 사용할 수 있으며, DNA Denaturation과 Bisulfite Conversion 과정은 Single-Tube Reaction에서 동시에 수행됩니다. 이후 Spin-Column 기반의 Desulphonation 및 Purification 과정을 거쳐 정제된 Bisulfite-Converted DNA를 PCR에 바로 사용할 수 있습니다.

PCR-Ready Bisulfite Converted DNA

Salmon sperm DNA were spiked with fully (CpG) methylated, linearlized pUC19 and pOX plasmid (1.5% w/w) and subjected to bisulfite conversion using the EZ DNA Methylation – Gold® kit. Two amplicons, 453 bp and 314 bp, were successfully amplified from the bisulfite converted pUC19 and pOX plasmid DNA, repectively. The amplicons were analyzed in a 2.0% (w/v) TAE/agarose/ EtBr gel. The size marker “M” is a 100 bp ladder.
